What is Motherwort Extract(Leonorus cardiaca)?

Motherwort (Leonurus cardiaca) is a herbaceous perennial plant in the mint family, Lamiaceae. Other common names include Throw-wort, Lion's Ear, and Lion's Tail. Lion's Tail also being a common name for Leonotis leonurus, and Lion's Ear, a common name for Leonotis nepetifolia. Originally from Central Asia it is now found worldwide, spread largely due to its use as a herbal remedy.

Motherwort has been used for hundreds of years, for women's issues. Wort is an Old English word meaning "to heal". Combine mother and wort, to see that it is aligned with the healing of a woman after childbirth and during her recovery. Motherwort is used for menstruation issues and nervous problems, and is said to be a gentle heart tonic. In fact, it is currently approved in Germany for nervous heart conditions, and as a treatment for Hypothyroidism.

Four new diterpenoids (1-4), along with eight known diterpenoids (5-12) were isolated from the acetone extract of Leonurus herb (aerial parts of Leonurus sibiricus L.). The structures of the compounds were determined by spectroscopic methods. Among the isolated compounds, compounds 2, 3, 8, 9 and 10 showed inhibitory activity against human liver cytosol estrogen sulfotransferase (E-ST), which plays a key role in the maintenance of cellular estrogen levels. Compound 2 showed the strongest activity with an IC50 value of 7.9 μM, which is comparable to the activity of the positive control, meclofenamic acid (IC50 5.4 μM).

Traditional Mongolian medicine (TMM) uses preparations from herbs as one form of medication for the treatment of a diversity of diseases including diabetes mellitus (DM). We evaluated the effect of extracts from the plant Leonurus sibiricus L. (LS), used in TMM to treat typical symptoms of type 2 DM, on insulin secretion, electrophysiological properties, intracellular calcium concentration and cell proliferation of INS-1E insulinoma cells under standard cell culture conditions (SCC; 11.1mM glucose).


Insulin secretion was measured by ELISA, electrical properties were assessed by whole cell patch clamping, intracellular calcium concentration (Cai) by Fluo-4 time lapse imaging, insulin receptor expression was verified by RT-PCR and cell proliferation assessed by CellTiter-Glo® cell viability assay.


Insulin released from INS-1E cells into the culture medium over 24h was significantly increased in presence of 500mg/L aqueous LS extract (LS OWE) as well as methanolic LS extract (LS MeOH/H2O) but not in the presence of the butanol-soluble extract (LS MeOH/BuOH). Acute application of LS OWE resulted in a depolarization of the cell membrane potential paralleled by an initial increase and subsequent decline and silencing of action potential frequency, by KATP channel inhibition, persisting depolarization and an increase in Cai. The electrophysiological effects were comparable to those of 100μM tolbutamide, which, however failed to elevate insulin secretion under SCC. Furthermore all LS extracts stimulated INS-1E cell proliferation.


The finding that extracts from Leonurus sibiricus L. enhance insulin secretion and/or foster cell proliferation may provide possible explanations for the underlying therapeutic principles in the empirical use of LS-containing formulations in DM and DM-related disorders as applied in TMM.

We report an efficient and powerful green process to enhance the antibacterial activities of the Leonuri herba extract. Plant sources, especially leaves and herbs, are precious for the generation of gold and silver nanoparticles. Various kinds of polyphenols and hydroxyl groups are capable of processing a reduction reaction to generate metals from its corresponding salts. We have prepared gold and silver nanoparticles with 70% ethanol and water extracts. No other toxic chemicals were utilized and the extracts played dual roles as reducing and stabilizing agents. For the generation of nanoparticles, both oven incubation and autoclaving methods were applied and the reaction conditions were optimized. Surface plasmon resonance band indicated that the formation of nanoparticles was successful. Images of high-resolution transmission electron microscopy revealed mostly spherical nanoparticles ranging from 9.9 to 13.0 nm in size. A water extract containing silver nanoparticles exhibited remarkable (approximately 127-fold) enhancement in antibacterial activities against Pseudomonas aeruginosa, Escherichia coli and Enterobacter cloacae when compared with the water extract alone. In addition, antibacterial activity towards Gram-negative bacteria was greater than that against Gram-positive bacteria. The process reported here has the potential to be a new approach to improve the antibacterial activities of plant extracts.

The neuronal damage following cerebral ischemia is a serious risk to stroke patients. The aim of this study was to investigate the neuroprotective effects of alkaloid extract from Leonurus heterophyllus (LHAE) on cerebral ischemic injury. After 24 h of reperfusion following ischemia for 2 h induced by middle cerebral artery occlusion (MCAO), some rats were intraperitoneally administered different doses of LHAE (3.6, 7.2, 14.4 mg/kg, respectively). Neurological examination was measured in all animals. Infarct volume, myeloperoxidase (MPO) activity, levels of nitrate/nitrite metabolite (NO) and apoptosis ratio of nerve fiber in brain were determined. The results showed that LHAE at 7.2 mg/kg or 14.4 mg/kg exerted significantly decreasing neurological deficit scores and reducing the infarct volume on rats with focal cerebral ischemic injury (p<0.05). At those dose, the MPO content were significantly decreased in ischemic brain as compared with model group (p<0.05). LHAE at 14.4 mg/kg significantly decreased the NO level compared with the model group (p<0.05). In addition, LHAE significantly decreased the apoptosis ratio of nerve fiber compared with the model group (p<0.05). This study suggests that LHAE may be used for treatment of ischemic stroke as a neuroprotective agent. Further studies are warranted to assess the efficacy and safety of LHAE in patients.

In the course of screening plants used in natural medicines as memory enhancers, a 70% ethanol extract of the aerial parts of Leonurus heterophyllus showed significant AChE inhibitory activity. Bioassay-guided fractionation and repeated column chromatography led to the isolation of a new labdane-type diterpenoids (1), named leoheteronin F, and six known compounds (2-7). The chemical structures of isolated compounds were elucidated based on extensive 1D and 2D NMR spectroscopic data. The isolates 1-7 were investigated in vitro for their anticholinesterase activity using mouse cortex AChE enzyme. Leoheteronin A (5) and leopersin G (7), which possess a 15,16-epoxy group at the side chain, were found to be potent in the inhibition of AChE.

Leonurus cardiaca L. (Lamiaceae) is used traditionally for its sedative, hypotensive and cardiotonic effects. Due to the lack of clinical data regarding its effect in patients, a study was carried out to assess the clinical efficacy of Leonurus oil extract (LOE) in patients with arterial hypertension stages 1 and 2, accompanied by anxiety and sleep disorders. Fifty patients were treated for 28 days with 1200 mg LOE per day. Positive effects of LOE on psycho-emotional status and arterial blood pressure in patients with stage 1 hypertension were observed 1 week earlier than in patients with stage 2 hypertension. According to the Clinical Global Impression (CGI) scale, a significant improvement in the symptoms of anxiety and depression was observed in 32% of patients, a moderate improvement in 48% and a weak effect in 8%; 12% of patients did not respond to therapy. Side effects were minimal in all groups. Leonurus oil extract may therefore be a potentially effective therapeutic agent for patients with arterial hypertension and concurrent psycho-neurological disorders.

Side effects and safety of Motherwort Extract(Leonorus cardiaca)

Motherwort is POSSIBLY SAFE for most people. Motherwort contains alkaloid stachydrine. Because of its ability to stimulate uterine contractions, it is not recommended during pregnancy, except during the last few days, when it is used to stimulate labor.
