What is Yeast Extract?

Yeast is a single cell micro-organism (fungus) that converts sugar and starch into carbon dioxide bubbles and alcohol.

A common method for making yeast extract for supplements or food products, such as Vegemite and Marmite, is to add salt to a suspension of yeast. The yeast cells shrivel up and start to break down, especially once heat is added. The thick cell walls of the yeast are then removed, which leaves the nutrient-rich contents of the cells. The processed extract is a very good source of protein, vitamins and minerals.

How does work Yeast Extract Beta Glucan?

In one research effort, it was found that yeast (1,3) Beta glucan binds directly to the Dectin-1 receptor on macrophages, giving us our first line of defense against dangerous bacteria and other health challenges. Dectin-1 is believed to be how Beta glucan is taken up into the body in the intestinal tract, as well as how it activates the macrophage to do its job as a molecular scavenger. This would make yeast Beta glucan extremely effective in helping the body fight off infection and/or help macrophages clear out abscesses or dead cells.

There is another receptor, the CR3 receptor on leukocytes, which helps fight infections and tumors by binding to antibodies along with Beta glucan in such a way that both must be bound for an effect to take place. When Beta glucan and the antibody attach to the CR3 receptor on leukocytes, the result is a destruction of a particular pathogen or tumor cell.

Treatment of hot water extract of the sclerotium of Polyporus rhinocerus (PRW) with murine macrophages including RAW 264.7 cell line and primary macrophages (PMs) could enhance their functional activities. These include a significant up-regulation of pinocytosis; an increase in the production of reactive oxygen species (ROS) and nitric oxide (NO); an increase in tumor necrosis factor alpha (TNF-alpha) production and inducible nitric oxide synthase (iNOS) expression in both RAW 264.7 cells and PMs. Cell surface receptors for yeast-derived beta-glucan, including Dectin-1, CR3, and TLR2, were determined by flow cytometry, and the expression of Dectin-1+ cells on the cell surface decreased in the responses of PMs to PRW. PRW increased phosphorylation of IkappaBalpha, which could trigger the nuclear factor kappa B (NF-kappaB) signal pathway for macrophage activation in RAW 264.7 cells. Therefore, the immunomodulatory effect of PRW could be mediated by macrophage activation via the NF-kappaB signal pathway.

Pathogens use a variety of strategies to evade host immune defenses. A powerful way to suppress immune function is to increase intracellular concentrations of cAMP in host immune cells, which dampens inflammatory responses and prevents microbial killing. We found that the yeast cell wall extract, zymosan, is capable of increasing intracellular cAMP and activates the protein kinase A pathway in bone marrow derived macrophage (BMDM) cells from mice. This response is dependent on adenylyl cyclase type VII (AC7) and heterotrimeric G proteins, primarily G(12/13). Consequently, zymosan induced production of the inflammatory cytokine, TNFα, was much stronger in BMDMs from AC7 deficient mice compared to the response in wild type cells. In a model of zymosan induced peritonitis, mice deficient of AC7 in the myeloid lineage displayed prolonged inflammation. We propose that zymosan induced increases in cAMP and activation of PKA serve as a mechanism to dampen inflammatory responses in host cells, which consequently favors the survival of microbes. This would also help explain a well documented phenomenon, that the ability of zymosan to stimulate inflammatory cytokine responses via TLR2 appears to be weaker than other stimuli of TLR2.
